Sessegnon, 19, arrives at Tottenham to provide competition for Danny Rose and Ben Davies at left-back, although he is also comfortable playing on the left side of midfield and in attack.
Spurs academy prospect Josh Onomah has moved in the other direction as part of the deal.
Onomah, 22, spent last season on loan at Sheffield Wednesday where he made 15 appearances in the Championship.
Sessegnon was first linked with a move away from Fulham after he scored 16 Championship goals as a 17-year-old to help the club earn promotion to the Premier League in 2017.
But despite interest from Spurs and manager Mauricio Pochettino, among other top Premier League sides,
Sessegnon remained at his hometown club and made 26 Premier League appearances last season, scoring two goals, as Fulham were relegated back to the Championship.
Sessegnon, who has made 10 appearances for England U21s, joins midfielder Tanguy Ndombele and Jack Clarke in joining Spurs this summer.
